 +====== Plugin FIGARO: 3D PSF estimation for optical microscopy ======

 +This plugin FIGARO helps tracking resolution of microscope systems by extracting dimensions and orientation of standardized microbeads images, acquired from test samples. In the development of optical microscopes for biomedical imaging, the evaluation of resolution is a fundamental parameter achieved by Point Spread Function (PSF) measurements. Sometimes, __**PSF measurement**__ procedure is not easy or impossible in case of **microspheres images presenting a high noise level**. The current method proposed into the plugin FIGARO is based on a variational approach for PSF modeling through multivariate Gaussian fitting, adapted to images ​ acquired in a high noise context. ​

 +  A- Before running ​
 +   1- Record a 3D image of a slice of fluorescent microbeads
 +   2- Crop the 3D image of a single microbead from the native image
 +   3- Load the image of the single 3D microbead in Fiji (or ImageJ)
 +   4- Run the plugin FIGARO; a specific window of inputs appears
 +   5- Inquire the image parameters
 +       - pixel size in x, y and z direction named dx, dy and dz
 +       - excitation wavelength in case of a multiphoton microscope

 +       - Numerical aperture ​

 +===== Installation =====

 +In order to use FIGARO plugin, please, download FIGARO_-1.0.jar and place it into the folder of Fiji named "​plugin"​. Restart Fiji. The plugin FIGARO appears into the plugin list of Fiji, gathering all the plugins already installed. Then, load the 3D images of a unique microsphere. Enter the pixel sizes in the 3 dimensions x, y and z. Excitation wavelength, numerical apearture, index and bead size can be filled. These information are involved in the generation of the PSF report for comparing the estimated PSF value with theoretical optimal value which can be deduced.
